GENOMIC ANALYSIS AND COMPARISON OF VERY VIRULENT INFECTIOUS BURSAL DISEASE VIRUS (VVIBDV) AFFECTING MALAYSIAN POULTRY CHICKENS WITH OTHER IBDV STRAINS

Virus-like particles (VLPs) are empty virus particles consisting of typically, the outer virus capsid proteins. They do not contain native viral nucleic acids, hence they are non-infectious and highly immunogenic. Many viruses have been reported to form VLPs when only structural envelope proteins are expressed in host cells. Expression of infectious busal disease virus (IBDV) polyprotein containing two structural envelope proteins was also shown to lead to the formation of VLPs. So far, IBDV VLPs formation had been reported in mammalian, insect and bacterial cells but not in yeast cells. The proposed study intends to find out whether IBDV VLPs could be formed in yeast cells. IBDV is the causative agent of an economically important disease affecting juvenile chickens (Gallus gallus) which results in high mortalility and morbidity of the animals. As this study will produce VLPs in yeast cells, the generated VLPs will later have the potential to be used as a subunit vaccine against IBDV infection and possible for large scale production. To form these VLPs, transformation of Pichia pastoris with a vector carrying segment A viral genome will be conducted. The VLPs formation is detected by observation of yeast cell lysates under electron microscope. In short, the proposed study will be beneficial for the poultry industry as it leads to the formation of subunit vaccine that protects chicken from infectious bursal disease and it will also add to the scientific knowledge for possibility of IBDV VLPs formation in yeast cells.
